Abstract

High-Molecular-weight (HMW) DNA from red blood cells of a female peking duck was prepared by embedding in low-melting-point (LMP) agarose. Performed a double-size selection for partially digested genomic DNA. BAC-DNA prepared from randomly selected white clones of each transformation digested with NotI restriction enzyme and subjected to pulsed-field gel electrophoresis to determine the size of the genomic insert fragments. Optimized the conditions for the construction of duck BAC library, the average size of inserts is up to 106 kb, ratio of non-insertion is about 2%, the library to be approximate 3-4 fold genome coverage.
